Abstract In the in vitro propagation system, zygotic embryos have been used frequently for high propagation efficiency, although these explants often bring morphological and genetic variation. As it was observed that shoots were developed in vitro both from the embryos and endosperms of the immature seeds of Xanthoceras sorbifolium, polyploidy and DNA variation was investigated for those propagules. In DNA content 2C occupied 70%, while 4C 15%~17%. In the variation study using RAPD markers, no differences were found among propagules and between propagules and seedlings. It suggests that this propagation system can provide the in vitro plants stable in genetic constitution.
